Summary Somatic embryos produced in vitro may exhibit structural abnormalities that affect their subsequent germination and conversion into plants. To assess the influence of auxin type on embryo initiation and development, a morphological and histological comparison was made of pecan (Carya illinoinensis) somatic embryogenic cultures induced on media with naphthaleneacetic acid or 2,4-dichlorophenoxyacetic acid (2,4-D), using light and scanning electron microscopy. Both auxins promoted enhanced cell division, particularly in subepidermal cell layers. However, notable differences were observed in mitotic activity, location of embryogenic cell proliferation, epidermal continuity, callus growth, and embryo morphology. Cultures induced on naphthaleneacetic acid had embryogenic regions composed of homogeneous, isodiametric, meristematic cells. Embryos derived from these cultures generally had a normal morphology, were single, and had a discrete apical meristem. In contrast, tissues induced on media with 2,4-D had more intense and heterogeneous regions of cell division. Proliferating cell regions were composed of meristematic cells interspersed with callus and involved more extensive regions of the mesophyll. Marked callus proliferation caused epidermal rupture in some areas. Embryos induced on medium with 2,4-D had a higher incidence of abnormalities that included fasciated, fan-shaped, and tubular embryos. Defined apical meristems were often lacking or partially obliterated due to callus proliferation. 对野生湖南山核桃果仁的油脂及其组分、总糖、蛋白质及其氨基酸组成、维生素E、维生素B1、钙、铁、锌、铜、钼等元素含量进行分析.果仁中粗脂肪、总糖、粗蛋白质、维生素E、维生素B1的含量分别为56.57%、3.51%、8.18%、3.06 mg/100 g和1.19 mg/100 g;钙、铁、锌、铜、钼元素含量的含量分别为114.6 mg/100 g、3.26 mg/100g、5.14 mg/100g、1.69 mg/100g和0.12 mg/100g.果仁的脂肪主要由软脂酸、油酸,亚油酸、亚麻酸、花生四烯酸组成,其含量分别为6.41%,71.80%,19.28%,1.82%和0.69%;果仁中蛋白质氨基酸有16种,其中9种人体必需氨基酸都含有.  相似文献

Aggregates of globular and pre-globular stage somatic embryos from suspension cultures of pecan (Carya illinoensis Koch) were cultured on solidified media for embryo development. Embryo aggregates and pre-globular stage embryo masses were given various treatments to further ontologic development. A 2- to 4-wk mild dehydration of the embryo aggregates suppressed recurrent embryogenesis, promoted development of globular embryos into cotyledonary stage embryos, and enhanced plant development beyond germination. Fine embryogenic tissue masses filtered from suspension formed cotyledonary-staged embryos when the collection filters were plated on solified medium. The embryogenic capacity of preglobular stage embryo masses was compared between media supplemented with varying concentrations of polyethylene glycol (molecular weight 8 000) vs. filter overlays. The filter paper overlays were not necessary for embryo development. An inverse relationship was found between the number of embryos that developed and the concentration of polyethylene glycol in the medium. However, this relationship was reversed for ability of embryos to germinate and develop into a plant.  相似文献

Host‐associated differentiation (HAD) is the formation of genetically distinct, host‐associated populations created and maintained by ecologically mediated reproductive isolation. HAD potentially accounts for species origins in parasites, including herbivorous insects. Although case studies testing the occurrence of HAD are accumulating, it is still unclear how common HAD is and which specific ecological traits explain its occurrence. To address these issues, studies are needed that include negative results (i.e., instances in which parasite populations do not exhibit HAD) and test for HAD across communities (i.e., several parasite species on the same set of host species). In this study, HAD was tested in a community of six species of Aphidomorpha (Hemiptera) that share a host‐plant pair: pecan [Carya illinoinensis (Wangenh.) K.Koch] and water hickory [Carya aquatica (F.Michx) Nutt., both Juglandaceae] trees. All six species are parthenogenetic and three species are endophagous, traits that can exacerbate host‐specific selection. AFLP markers were employed to detect the presence of genetically distinct, host‐associated populations for each insect species. Strict HAD (i.e., the occurrence of genetically distinct pecan‐associated and water hickory‐associated genotypes) was found in Phylloxera notabilis Pergande (Phylloxeridae), Phylloxera devastatrix Pergande, and Monelliopsis pecanis Bissel (Aphididae). Monellia caryella Fitch (Aphididae) displayed a pattern of partial HAD (i.e., the occurrence of only a genetically distinct pecan‐associated genotype). No HAD was found in Melanocallis caryaefoliae Davis (Aphididae) or Phylloxera texana Stoetzel. The pattern of HAD occurrence in the pecan and water hickory Aphidomorpha community indicated that neither parthenogenesis nor endophagy sufficiently explain the occurrence of HAD in this system.  相似文献

Pecan [Carya illinoensis (Wanngenh) K. Koch] is one of the most important nut crops in arid and semiarid regions of Mexico. Here, most pecans are grown in saline soils having poor permeability which are further degraded by the use of low-quality irrigation water. Salinity adversely affects both pecan nut quality and yield. Little work has been done to explore the physiological effects of salinity on native pecan trees. Here we examine physiological changes determined by exposure of pecan seedlings to sodium sulfate (Na2SO4) at four concentrations: 1000, 2000, 3000 and 4000 mg/L applied twice weekly over a 70 d period. Control plants were similarly irrigated but with water free of Na2SO4. The aim was to identify and quantify the putative salinity damage to native pecan seedlings growing in Chihuahua, Mexico. Seedlings exposed to Na2SO4 were of reduced height and stem diameter. At the highest exposure level (4000 mg/L), proline concentration in the leaflets was 820% higher (2.63 mg/g) than in the controls (0.32 mg/g), and chlorophyll was 35% lower (23.4 mg/L) than in the controls (36 mg/L). Meanwhile, sulfate ion concentration was increased by 104% from 84.47 to 172.5 mg/g. Root biomass decreased by 310% (from 30.5 to 9.5 g) and foliar biomass decreased by 260% (from 26.7 to 10 g). No disease symptoms were apparent in any seedlings suggesting that these changes were induced by Na2SO4 stress alone. Of the physiological parameters measured, proline, chlorophyll and sulfate ion concentration, as well as root and shoot biomasses were strongly affected by irrigation with Na2SO4 at concentrations of 2000 mg/L and above.  相似文献

薄壳山核桃(Carya illinoensis)富含酚类代谢物,具有显著的抗氧化活性。近年来,我国薄壳山核桃产业快速发展,种植面积迅速扩大。明确薄壳山核桃中的酚类代谢物组成及保健功效对于促进健康产业发展具有重要意义。迄今为止,从薄壳山核桃中已检测出酚类代谢物67个,其中包括单宁36个、类黄酮22个、酚酸9个,水解后检测出酚类代谢物苷元34个。该文主要对薄壳山核桃酚类代谢物的组成、提取方法、含量、药理活性及与采后贮藏特性的关系进行综述,以期为今后深入研究薄壳山核桃酚类代谢物与其生理活性的关系及挖掘薄壳山核桃酚类代谢物的保健价值提供依据。  相似文献

薄壳山核桃﹝Carya illinoinensis ( Wangenh.) K. Koch﹞为胡桃科( Juglandaceae)山核桃属( Carya Nutt.)植物,又名美国山核桃、长山核桃,是世界上著名的经济树种之一[1-2]。目前,主要采用本砧嫁接的方法培育薄壳山核桃良种嫁接苗,而培育优质实生苗作为砧木是该技术的关键环节之一,因此,了解其种子萌发特性极为重要。目前,对薄壳山核桃种子萌发特性的研究主要集中在植物激素催芽方面[3-5],而有关激素催芽过程中的适宜浸泡天数、种子吸胀过程中相关的酶活性变化等问题尚不明确。  相似文献
